get a micrometer, caliper or good ruler and count
Also get a magnifier so you can see well. With a caliper you can just set it to measure the distance from one thread to another one farther away, the farther the better. Then use your magnifer to count the number of threads within this distance. Now divide the first number into the second number and the result will be threads per inch.
